What are peptides?

Peptides are  an integral part of most biologic processes and are present throughout every cell and tissue in the body. Maintenance of appropriate concentration and activity levels of peptides is necessary to achieve homeostasis and maintain health. The function of a peptide is determined by its size and amino acid sequence.

All peptides are composed of amino acids chemically linked together. Peptides range in size from two amino acids–dipeptides–to 50 amino acids. Polypeptides consist of 50-100 aminoacids. Proteins are made of 100's to thousands of amino acids. 

There are at least 700 different therapeutic peptides.

What are the different types of peptides?

Peptides can be:

  • Transporters-  like glucose transporters that are necessary for glucose to travel from the blood into the muscle
  • Enzymes - are biological catalysts that speed up metabolic reactions. Most of the hundreds of enzymes are peptides. 
  • Hormones- are biologic messengers that carry information from one tissue through the blood to a distant tissue. Two common classes of hormones are peptide and steroid hormones. Examples of peptide hormones include those involved in blood glucose regulation, such as insulin and glucagon, and those that regulate appetites, such as ghrelin and leptin.
  • Structural components - peptides like actin and myosin function as structural elements of the muscle and  some other peptides contribute to bone shape and strength.
  •  What are the most commonly Used Peptides?

    1. Ipomorelin: 

    Ipamorelin is the first selective GHRP (growth hormone releasing peptide) secretagogue and ghrelin receptor agonist.  Ipamorelin is one of the safest, most effective forms of growth hormone (GH) restoration, widely used to support age management, disease management, sports performance and GH deficiency treatment protocols, with virtually no side effects. It is considered “selective” since, unlike other GHRPs, this peptide doesn’t affect the release of cortisol, acetylcholine, prolactin or aldosterone, thereby minimizing side effects experienced with other GH therapies, such as increased hunger or irritability.

     Because there are virtually no negative side effects, Ipamorelin can be prescribed more aggressively and more frequently than other therapies without the risk of elevated cortisol or acetylcholine blood plasma levels. This helps optimize HGH levels for a longer period of time, leading to more successful health outcomes.  The physicians at Radiance recommend 8-12 week cycles depending on prior use and lifestyle factors.

    2. IPAMORELIN/CJC-1295

    CJC-1295 is a growth hormone releasing hormone (GHRH) analog of 29 amino acids. CJC-1295 has been shown to increase human growth hormone (HGH) as well as insulin-like growth factor (IGF-1) secretion and has been able to do that in large amounts.  CJC-1295 also works without increasing prolactin, leading to exponential fat loss and increased protein synthesis/muscle mass without negative side effects. CJC-1295 is shown to outperform Sermorelin (older peptide which is a GHRH as well) and is now surpassing its use, especially since the half life is longer.  

    From our special compounding pharmacies we are able to offer Ipamorelin/CJC-1295 combination peptide that is a combination of both a GHRH and a GHRP to act together on the pituitary and hypothalamus to exponentially increase levels of growth hormone in your body.  Researchers have found the combination can increase GH over 5x than when either were administered alone. The combination allows for maximum release of GH because the CJC- 1295 and Ipamorelin have different mechanisms of actions and work on different receptors (GHRH-R and Ghrelin-R) which helps to:

    3. BPC-157

    BPC-157 has shown impressive results with regards to healing and muscle growth.  BPC-157 (pentadecapeptide) was originally formulated to be an injectable peptide developed for alleviating stomach ulcers. BPC-157 is a sequence of body protection compounds formed of 15 amino acid chains. It has since been found to accelerate the healing of a huge variety of wounds in different areas such as skin, cornea, muscle, colon, blood vessels, bone and tendons. This peptide has many remarkable healing properties including granulation tissue formation (the onset of healing), angiogenesis (formation of new blood vessels) and production of collagen (a protein that provides structural support, strength, and elasticity to the human body).

     Scientists aren't certain about the exact mechanism BPC-157 achieves its healing powers, however, the hypotheses involve the regulation of growth factors like fibroblasts and other morphogenetic proteins that make bone, cartilage and tissue.  Its’ proangiogenic effects, and modulation of nitric oxide synthesis.

    What are some of the other useful Peptides?
  • 1. IGF-1.  is in the peptide family which has the same structure and size as insulin. It is a highly anabolic hormone that is released in the liver as well as in the peripheral tissues naturally so new muscle can be built. The main work of IGF-1 is to build new muscle tissues by promoting nitrogen retention and protein synthesis.  Regular users claim that the results of Long R3IGF-1 are leaner, harder, fuller muscular growth that does not dissipate like the size and strength gains from anabolic steroids.  IGF-1 not only makes muscle fibers bigger, but also makes more of them. IGF-1 has potent effect on fat metabolism and helps the body to burn fat rapidly. It also improves mental functions such as reflexes and learning ability.
  • IGF-1 has a very sensitive and unstable property with a very short half-life of only 10 to 20 minutes. A substitute of IGF-1, referred to as Long R3 IGF-1 or IGF-1 LR3, is usually used.  IGF-1 LR3 has longer life of 20 - 24 hours  so that the users can administer smaller amounts and does not have to use it as frequently.      2. DSIP

    DSIP is delta sleep inducing peptide, which means it promotes deep sleep in chronic insomniacs. Studies show that first injection produces normal sleep pattern, without any cognitive impairments.  In a placebo controlled double blind study with polysomnogram monitoring and psychological testing DSIP injection substantially improved night sleep with the first and additionally with repeated doses. These effects were maintained for the first posttreatment (placebo) night. Efficiency of night sleep and daytime rest reached the levels of normal controls.

    Two other studies showed improvement of sleep following a single injection of 25 nmol/kg of body weight. Repeated administration showed a build up with normalization of sleep pattern and structure after four administrations. Dosage: 250 mcg. S.C about 2 h before sleep. Some patients will need doses as high as 300-400 mcg.

    In addition to sleep, this peptide also has been observed to affect (Grat & Katin, 1984) electrophysiological activity, neurotransmitter levels in the brain, circadian and locomotor patterns, hormonal levels, psychological performance, and the activity of neuropharmacological drugs including their withdrawal.

    DSIP works as well in people dependent on benzodiazepines and a few studies have revealed that this peptide may be ideally used to combat any withdrawal symptoms from the chronic use of sleeping pills and normalization of sleep pattern. It may also be useful in chronic pain.


    3. PT-141 (Bremelanotide)

    It is a synthetic cyclic heptapeptide, a melanocortin receptor agonist, mainly acting as an MC3 and MC4 receptor agonist. It is an active metabolite of Melanotan-II. It is also called “libido drug” by researchers and is a very powerful aphrodisiac.

    It works at the brain level, activating MC4 receptors, thus eliciting a rather natural sexual response. It increases the "pleasure hormone" dopamine as well.

    It is effective in both sexes, for Erectile Dysfunction (ED) in males and Female Arousal Dysfunction  or Hypoactive Sexual Desire Disorder in women.

    Elimination half-life of PT-141 is 120 min.

    Adverse effects described in the medical literature  include nausea, flushing, headache, injection-site pain and itchiness.

    Dosage: 1.25 mg initially; may be increased to 1.75 mg S/C if needed 2-4 h before sexual intercourse.

    4.  AOD-9604:

    AOD-9604 is based on a small part of the human growth hormone molecule. This hormone, which occurs naturally in the body and which stimulates fat metabolism, is suppressed in obese people.

    This peptide, which stimulates the metabolism of body fat, is the first of its kind. All other obesity drugs artificially reduce appetite or food absorption.

    This peptide was taken orally once daily by 300 obese patients at five trial sites over a 12-week period. Six doses were used – 0 mg (placebo), 1mg, 5 mg, 10 mg, 20 mg and 30 mg.

    The group receiving the 1mg dose lost the most weight, averaging a weight loss over the 12 weeks of 2.8 Kg, more than triple the weight lost by those on placebo, who lost an average of 0.8 Kg. The rate of weight loss was maintained throughout the treatment period, an encouraging trend for expectations of longer-term dosing.

    The weight lost by the 1mg group was slightly more than that achieved by the world’s largest selling prescription obesity medication in similar trials over the same period, without its troublesome side effects. The trial results also demonstrated a small but consistent improvement in cholesterol profiles, and a reduction in the number of patients with impaired glucose tolerance.

    Obesity is the most common health problem, and has reached epidemic proportions according to the World Health Organisation. More than 20% of the adult population in developed countries are obese – more than 300 million adults worldwide. In addition, more than 50% of adults in developed countries are overweight. Obesity is associated with other health-related problems such as heart disease, cancer and diabetes.

    5. Kisspeptin

    Kisspeptin (formerly known as metastin) is a protein that is encoded by the KISS1 gene in humans.It is a G-protein coupled receptor ligand for GPR54. Kiss1 was originally identified as a human metastasis suppressor gene that has the ability to suppress melanoma and breast cancer metastasis.

    KISS1 generates a c-terminally amidated peptide, metastin which functions as the endogenous ligand of the g-protein coupled receptor gpr54. Activation of the receptor inhibits cell proliferation and cell migration, key characteristics of tumor metastasis.

    KISS1 is a metastasis suppressor protein that suppresses metastases of melanomas and breast carcinomas without affecting tumorigenicity.KISS1 functions to inhibit chemotaxis and invasion, attenuating metastasis in malignant melanomas. KISS1 is involved in the regulation of cell-matrix adhesion. Expression of KiSS1 is localized to specific regions of the hypothalamus in numerous species and is regulated by gonadal steroids and across the estrous cycle.
